Low molecular mass GTP-binding proteins in subcellular fractions of the pancreas: regulated... teins are present on adrenal chromaffin granules. Matsuoka and Dolly (29) have reported localized to synaptic vesicles, and Fisher von Mollard et al. (15) have identified one these as rab3A. How these function in regulated exocytosis is not apparent at this time. However, analysis a secretory mutant in yeast has shown that constitutive secretion in yeast requires the sec4 gene product, which is a protein found in both cytosolic and membrane-associated forms (36). In sec4 mutants, secretory vesicles accumulate in the cytosol, suggesting some step between the trans-Golgi and plasma membrane fusion is aberrant. Other recent studies have demonstrated that may also be phosphorylated (23, 26), and it is suggested that phosphorylation GTPbinding would be a possible regulatory mechanism. Regulation protein phosphorylation by secretagogueshas been demonstrated convincingly in pancreatic acinar cells (7, 8, 12, 19), although the nature most the regulated has not been established. We, therefore, mapped the total and subcellular distribution an exocrine secretory cell, the pancreatic acinar cell, using high-resolution twodimensional gel electrophoresis and compared it with the pattern labeled with 32P04.
