The forward primer was 5'-AGGATGAA- GTCTGTAAGGATAGGCAGAGCA-3' and the reverse primer was 5'-TTCTCTGTGTAGTCCTGGCTGTCCTGGAA-3'. Allele detection: A strain difference was detected by PCR. The PCR conditions consisted of a predenaturation step at 94~ for 2 min, followed by 30 cycles at 94~ for 45 s, 59~ for 1 min, and 72~ for 1 rain. The PCR products were about 200 bp and 250 bp in the C57BL/6J and M. spretus DNAs, respectively. Previously identified homologs: rat CA V [4]; human CA V[5]. Discussion: The mouse Car5 gene encodes a 298-amino acid pre- cursor prepropeptide whose 28-amino acid leader sequence is pro- teolytically cleaved to produce a mature 270 amino acid CA V protein localized to the mitochondrial matrix [4].
