G8 Factors Regulating the Immune Response type has been defined as a central failure of responsiveness brought about by exposure to antigen, immunologically competent cells becoming unable to initiate synthesis of a restricted range of antibodies (or comparable recognition units for cellular immunity). An essential feature of this definition is central failure, cells becoming unable to respond; there is no failure of antigen to react them (afferent block) or secretion of a product that is subsequently blocked (efferent block). Another essential feature is specificity, for only a restricted range of capabilities are lost; this is in contrast to the non-specific unrespon- sivencss that can be produced with radiation or immunosuppressive drugs. Unresponsiveness induced by antigen is of direct interest because of the hope that the induction of tolerance will in the future aid organ transplantation. It is also to be hoped that study of the termination of tolerance will reveal autoimmune disease mechanisms.
