Abstract

•SYBR Green I-based real-time RT-PCR in combination with melting curve analysis (MCA) was optimized for the detection and discrimination of grapevine viruses.•The assay was evaluated to confirm the feasibility of this technique in the detection of grapevine viruses from field samples and insect vectors.•Besides virus diagnostics, the SYBR Green I-based real-time RT-PCR MCA assay provides a superior resolution power to examine competition dynamics among frequently co-occurring viruses in grapevines and explore the consequences of co-infections on virus-host interactions and spread of individual viruses.
