•In 13 lung cancer cell lines, with qPCR BRS-3mRNA was expressed in 92%, GRPR and NMBR in 85%.•In 54% of lung cancer cell lines BRS-3mRNA was present in larger amounts than GRPR/NMBR.•Positive BRS-3 cell lines in 100% showed either/or MAPK/PLC activation with a BRS-3 agonist.•Selective BRS-3 agonists stimulated growth of BRS-3 positive lung cancer cell lines.•In 54% of cell-lines BRS-3 agonists stimulated growth was mediated by EGFR transactivation.
