•Developed a novel and rapid (20min) RT-exoRPA assay for the detection of Rose rosette virus (RRV).•The RT-exoRPA primers/probe was highly specific to RRV.•The assay was highly sensitive, detecting up to 1 fg of virus.•A rapid viral RNA extraction procedure for RRV (<5min) was developed from different tissue sources.•The assay detected RRV from different tissue sources of infected plants, as well as from different rose varieties.
