Tissue Typing Using a Routine One‐Step Lymphocyte Separation Procedure Summary. A reliable rapid one‐step method is described for the preparation of lymphocytes from 0.25 ml aliquots of defibrinated whole venous blood for tissue typing. An average yield of 70% lymphocytes is obtained relatively free from platelets, erythrocytes and granulocytes even in the presence of large numbers of granulocytes in blood obtained from cadaver donors. Using a high‐speed micro‐centrifuge lymphocytes can be prepared from defibrinated blood within 10 min and it is thus possible to complete a tissue typing test within 75 min.
