Rapid effect on suckling of an oxytocin antisense oligonucleotide administered into rat supraoptic nucleus

Rapid effect on suckling of an oxytocin antisense oligonucleotide administered into rat... thought to be due to hybridizati of the oligucleotides with mRNA coding for the respective peptide and csequent inhibiti of its synthesis. The role of the napeptide oxytocin (Oxt), which is synthesized within the hypothalamic supraoptic (S) and paraventricular nuclei, during in the lactating rat is well established. It is released into the bloodstream from neurohypophysial terminals during each milk ejecti reflex (8), as well as within the magnocellular nuclei themselves (16, 18), where it further stimulates the activity of Oxt neurs (10, 17). In the present study we have used the cscious lactating rat to study of an Oxt oligucleotide infused bilaterally into the S -related parameters (milk transfer to pups and number of milk ejecti reflexes during a 30-min period; plasma Oxt levels). Furthermore, Oxt ctent of the posterior pituitary and Oxt immunoreactivity within the hypothalamus were estimated to determine whether the neuroendocrine respses observed during were due to depleted stores of Oxt within the hypothalamo-neurohypophysial system. MATERIALS AND METHODS IT IS NOW KNOWN that mRNA and DNA oligo- nucleotides can be injected into the brain, where they are taken up by neurs (13, 26) to alter physiological and behavioral respses (1, 14, 15, 25). oligucleotides http://www.deepdyve.com/assets/images/DeepDyve-Logo-lg.png AJP - Regulatory, Integrative and Comparative Physiology The American Physiological Society
