To elucidate the potential role of the T lymphocyte, we investigated the effects of interferon gamma (INF), a product of activated lymphocytes, on smooth muscle cell proliferation. METHODS Human arterial smooth muscle cells (HUA-SMCs) were prepared from human um- bilical artery by the method of R. Ross and were maintained in RPMI 1640 medium supplemented with 20% FCS, 10 ng/ml basic FGF. The third passage of HUA-SMCs was seeded in a 24-well tissue culture dish (2 cm*/well) and cultured in regular medium with or without human recombinant interferon gamma (provided by Takeda Pharmaceutical Co.) for 3-5 days. Cells were trypsinized with 0.25% trypsin/EDTA and the cell number was determined with a Coulter particle counter. RESULTS AND DISCUSSION Under our experimental conditions, HUA-SMCs grew logarithmically for 12-1 5 days and became confluent approximately at 2 x 10’ cells/cm2.
