TY - JOUR AU - Schluederberg, Ann AB - Structures with the dimension and morphology of Dane particle cores were isolated and concentrated by a simple one-step procedure from human serum containing hepatitis B antigen. The method involves centrifugation of serum layered over a density gradient containing a zone of Nonidet-P40. Cores found at a solution density of 1.29–1.30 g/ml in Ficoll-sucrose gradients prepared in D2O reacted, in immunodiffusion tests, with a human serum containing core antibody. A modification of the method yielded partially degraded Dane particles in a highly aggregated array that revealed a previously undescribed outer morphology. Cores were surrounded by an areola containing regularly spaced projections 14 nm long that appeared to originate at individual sites on the surface of the core.
