AB - A rapid and facile procedure has been developed for preparing the desired acetylene concentration in large sealed chambers for acetylene reduction assays. The gas dilution system incorporated an inexpensive manometer to monitor evacuation of a known amount of air from the chamber. A simple valving system then allowed for easy injection of acetylene to produce the desired assay conditions. Although 34‐L chambers were used, the procedure is amenable to sealed chambers of any size. The entire procedure takes <1 min and exhibits excellent precision. This technique facilitates the use of very large assay chambers, and resolves the cumbersome problems and time involved in handling large volumes of gas.
