IMMUNOLOGICAL DIFFERENTIATION BETWEEN BURSAPHELENCHUS XYLOPHILUS AND B. MUCRONATUS BY C. LAWLER1) P. JOYCE2) and M. A. HARMEY1) 1) Dept. of Botany, University College Dublin, Dublin 4, Ireland; 2) Zoology Dept., University College Dublin Antisera to Bursaphelenchus xylophilus was prepared by two different immunisation protocols. The antibodies were examined using the Enzyme Linked Immunoabsorbent Assay (ELISA) and West- ern Blots. The polyclonal antibodies obtained by the different immunisation schedules did not clearly distinguish B. xylophilus from B. mucronatus using the ELISA system. The use of rats immunotolerised to B. mucronatus antigen produced polyclonal antibodies which could distinguish specific antigens of certain B. xylofihilus populations when examined using Western Blots. The banding patterns of the B. xylophilus isolates examined using the polyclonal antibodies obtained from immunotolerised rats indicate that Japanese and United States populations are more closely related to each other than to the Canadian populations.
