After thickly greasing the outside surface of the cylinder with silicon paste, a glass cover with a bore-hole (i.d. 1 cm) is put on, which contains a sealed-in connecting pipe closed with aluminium foil. Its distance from the inside wall of the cylinder is ca 1.5 cm, and its inner diameter is ca 1.0 cm. An impermeable connection to the glass container is obtained by pressing the cover and turning it around its centre. The container with the soil added is autoclaved three times during 30 minutes at 1.5 atm in intervals off 24 hours. Then, surface-sterilized seeds are dropped under sterile conditions through the connecting pipe into the cylinder and the system is closed by a sterile glass bung which is also sealed with silicon paste. A circular dispersion of the seeds can be obtained by carefully turning the cover around its centre. The lower part of the container is covered with aluminium foil to the level of the soil surface to exclude light.
